Antimicrobial dressing efficacy against mature Pseudomonas aeruginosa biofilm on porcine skin explants

چکیده

Methods: The assessments were performed on biofilm attached to dermal tissue using an ex vivo porcine skin explant biofilm model. Its use as a dermal substrate for attachment and the primary source of nutrition provides an alternative cost-effective method that more closely mimics in vivo conditions, with the advantages of an in vitro system, to assess the antimicrobial efficacy of treatments against mature biofilms. This ex vivo porcine skin explant biofilm model has been further validated against data generated from an in vivo pig burn wound model that was used to assess the efficacy of silver dressings against PAO1 biofilms. Five types of antimicrobial agents and four types of moisture dressings were assessed.

Antimicrobial dressing efficacy against mature Pseudomonas aeruginosa biofilm on porcine skin explants.

An ex vivo porcine skin explant biofilm model that preserves key properties of biofilm attached to skin at different levels of maturity (0-3 days) was used to assess the efficacy of commercially available antimicrobial dressings and topical treatments. Assays were also performed on the subpopulation of antibiotic tolerant biofilm generated by 24 hours of pre-treatment with gentamicin (120× mini...

Impact of a novel, antimicrobial dressing on in vivo, Pseudomonas aeruginosa wound biofilm: quantitative comparative analysis using a rabbit ear model.

The importance of bacterial biofilms to chronic wound pathogenesis is well established. Different treatment modalities, including topical dressings, have yet to show consistent efficacy against wound biofilm. This study evaluates the impact of a novel, antimicrobial Test Dressing on Pseudomonas aeruginosa biofilm-infected wounds. Six-mm dermal punch wounds in rabbit ears were inoculated with 10...

Efficient Eradication of Mature Pseudomonas aeruginosa Biofilm via Controlled Delivery of Nitric Oxide Combined with Antimicrobial Peptide and Antibiotics

Fast eradication of mature biofilms is the 'holy grail' in the clinical management of device-related infections. Endogenous nitric oxide (NO) produced by macrophages plays an important role in host defense against intracellular pathogens, and NO is a promising agent in preventing biofilms formation in vitro. However, the rate of delivery of NO by various NO donors (e.g., diazeniumdiolates, S-ni...

Study of antimicrobial effects of several antibiotics and iron oxide nanoparticles on biofilm producing pseudomonas aeruginosa

Objective(s): Pseudomonas aeruginosa is a nosocomial pathogen resistant to most antimicrobial treatments. Furthermore, it persists in adverse environments thereby forming biofilms on various surfaces. Researchers have therefore focused on antibiofilm strategies using nanoparticles due to their unique physicochemical properties. Superparamagnetic iron oxide nanoparticles (SIONPs) have recently s...
